Collection Instructions

Specimen:Blood, BAL and swab
Optimal Volume:2 mL blood or BAL; swab in 1 mL BD Universal Transport Media
Minimum\Peds Volume:1.2 mL blood; 0.5 mL BAL; or swab in 1 mL BD Universal Transport Media
Container:Purple (EDTA), BD Universal Transport Media (Plain syringe)
Collection Instructions:
Deliver to laboratory immediately. Specimen should be collected during acute phase of the disease as follows:
  • Blood: Collect in purple (EDTA) tube. Serum is acceptable but not preferred.
  • Eye, mouth or skin swab: Use swab to collect the specimen. Alternately transfer the sample (using fine sterile spatula) to Universal transport medium.
  • Genital swab: Rub genital lesion firmly with culture swab. Place in Universal transport medium.
  • Skin lesion: Using a tuberculin syringe, collect fluid from at least six vesicles. Place in Universal transport medium
 
Causes for Rejection:Feces and heparinized specimens, wound dressings


Processing and Shipping

Specimen Processing:Centrifuge and aliquot 1 mL, 0.5 mL minimum plasma/serum, bronchoalveolar lavage. Store in refrigerated or frozen. Use plastic storage container-no glass (will break on freezing). All swabs must be collected with flocked swab, refrigerate in 1 mL universal transport media (UTM).
Shipping Instructions:Ship serum/plasma or BAL refrigerated. Ship culturettes at refrigerated temperature.

Reference Range:

Negative

HSV 1&2 DNA not detected by PCR

Positive

HSV 1 DNA detected by PCR

HSV 2 DNA detected by PCR

A negative result does not rule out the presence of PCR inhibitors in the patient specimen or Herpes simplex virus DNA concentrations below the level of detection by the assay.

 

Use:Detection of HSV DNA and the ability to distinguish between different HSV types can be important for prognosis or for monitoring treatment success. This test should not be used to screen for an exposure to herpes simplex virus or ordered on asymptomatic patients.
