DNA Marker, Pre-BMT Engraftment Donor Testing
Abbrev Code: | DNENFT | ||
Order Code: | LAB4727 | Order Name: | Pre BMT Polymorphism Determination Donor |
Order Instructions: | For recipient samples, use code: RCENFT, LAB4728
Synonyms: | Bone Marrow Transplant DNA Marker; DNA Polymorphism; Molecular; Pre-Transplant Chimerism Study
Methodology: | PCR amplification of DNA and capillary electrophoresis
Test Includes: | BMT DNA processing, DNA marker pre-BMT polymorphism
Turnaround Time: | Performed Mon-Fri; results are reported 1-2 weeks after both donor and pre-BMT recipient specimens have been received..
Special Instructions: | Donor testing only needs to be done once for the same recipient. Duplicate donor testing will be canceled. Record the transplant recipient's name on request form when sending the donor specimen. See DNA Marker, Pre BMT Engraftment, Recipient Testing for additional information. |
Collection Instructions
Specimen: | Blood, cord blood, bone marrow aspirate, bone marrow biopsy, tissue biopsy or buccal scrapings |
Minimum\Peds Volume: | Contact laboratory if unable to obtain specified collection volume. |
Container: | Yellow (ACD, Solution A) tube available from laboratory Alternate Containers: Purple (EDTA) |
Collection Instructions: |
|
Causes for Rejection: | Frozen or clotted specimen; incorrect anticoagulant. DNA extracted at non-CLIA certified (or equivalent) lab.
Shared samples will not be accepted unless sent to Molecular first. Contamination will occur on automated hematology analyzers. |
Processing and Shipping
Specimen Processing: | Store blood, cord blood, bone marrow aspirate and buccal scrapings at room temperature. Store tissue at -70C. |
Shipping Instructions: | Ship blood, cord blood, bone marrow or buccal scrapings at room temperature. Swabs must arrive at testing laboratory within 24 hours and blood, bone marrow or cord blood within 5 days. Ship tissue at room temperature to arrive within 1 hour; alternatively, ship on dry ice to arrive within 24 hours, or send in RPMI at 4C. |
Stability: | Blood, bone marrow or cord blood: 5 days at room temperature.
Swabs: 24 hours at room temperature. Tissue: 1 hour at room temperature. Alternatively 24 hours at room temperature in RPMI, or frozen. |
Test Performed at or Referral Lab | Molecular Diagnostics (UMMC East Bank) |
Interpretive
Reference Range: | Results are reported as "Donor DNA processed for post-transplant engraftment polymorphism analysis." This does not indicate that a polymorphism was found between recipient and donor that will allow the lab to follow the DNA engraftment of patient post-bone marrow transplant. Each donor will have a separate report. Charges for donor testing will be billed under pre-BMT engraftment recipient. |
Use: | Evaluation for identification of informative polymorphic markers which are used to determine engraftment status subsequent to bone marrow transplantation. An informative marker is defined as one that demonstrates unique identity for recipient and donor DNA. |
