Collection Instructions

Specimen:Blood
Optimal Volume:10 mL and a 10 mL normal control from an unrelated donor
Minimum\Peds Volume:7 mL (adult) or 3 mL (pediatric) and 7 mL normal control from an unrelated donor
Container:Green (sodium heparin, no gel)
Alternate Containers: Yellow (ACD, Solution A) tube available from laboratory
Collection Instructions:Sample and control must be collected Monday through Thursday. Specimen and control must be in the original tubes and arrive at the reference laboratory within 48 hours of collection time and before noon on Friday. Testing is performed Tuesday through Friday only.
Patient Preparation:Do not order for patients younger than 3 months unless clinical history of candidiasis is present.
Causes for Rejection:Yellow (ACD Solution B); refrigerated or frozen samples; samples in transport longer than 48 hours.


Processing and Shipping

Specimen Processing:Whole blood in original container. Do not process. Send sample and control at room temperature.
Shipping Instructions:Ship at room temperature. Must arrive within 48 hours of testing and by noon on Friday. Live lymphocytes required. Critical ambient, do not refrigerate or freeze.

Interpretive

Reference Range:By report.
Use:
Phytohemagglutinin concanavalin A, pokeweed mitogen, Candida antigen and tetanus antigen are tested independently in lymphocyte culture. Lymphocyte proliferation in response to these mitogens and antigens is determined by 3H-thymidine incorporation.

Results are reported as counts per minute (CPM) mitogen or antigen stimulated versus a control culture and a Simulation Index (SI) which represents the ratio of CPM of the stimulated lymphocytes to the mean CPM of the unstimulated control.
