Please call the Molecular Diagnostics Lab before sending sample. Maternal engraftment patients Identify patient's name, mother's name, and sex of patient. Indicate "maternal engraftment", "donor engraftment", or "third party engraftment" on request form. "Donor engraftment" and "third party engraftment" require recipient and donor pre-transplant sample, and recipient post-transplant (or post-transfusion) sample.
Graft versus Host Patients For analysis to be completed, all three of the following sample types must be received: · Recipient cells (buccal swab) · Donor cells/DNA which have been previously banked (in HLA Lab or Cell Therapy) prior to transplant · Post-transplant material from patient

Causes for Rejection: | Clotted specimen; necrotic tissue, incorrect anticoagulant. Add on testing to hematology samples is not acceptable. DNA extracted at non-CLIA certified (or equivalent) lab. |
Contraindications: |
Blood is fractionated into CD3+ if WBC is greater than 5 x 109/L and sample is <48 hours old. Bone marrow is not fractionated. Results are reported with the exact percentage of donor engraftment. Sensitivity +5% indicates the minimum amount of control DNA marker detectable for this specimen.

Processing and Shipping
Specimen Processing: | Store blood and bone marrow aspirate at room temperature. Store tissue at -70 degrees C. Contact laboratory prior to shipping. |
Shipping Instructions: | Ship blood and bone marrow at room temperature to arrive within 24 hours. Ship tissue at room temperature to arrive within 1 hour; alternatively, ship on dry ice to arrive within 24 hours. |

Interpretive
Reference Range: |
Blood is fractionated into CD3+ and CD15+ if WBC is greater than 1.0 x 109/L and sample is less than 48 hours old. Bone marrow is not fractionated. Results are reported with the exact percentage of donor engraftment. “Sensitivity ±5%” indicates the minimum amount of control DNA marker detectable for this specimen.
When more than one of the ten STR markers is informative, the final result will be the average of all informative markers.
